谷氨酸（Glu）在酶的催化作用下可将NAD+还原成NADH，NADH在递氢物质的作用下将WST-8还原生成橙黄色的物质，该物质在450 nm有最大吸收峰。通过测定450 nm下吸光值可计算样本中Glu含量。
