检测原理 |
Gln在酶的催化作用下反应,同时NAD+被还原成NADH,NADH在递氢物质的作用下将WST-8还原生成橙黄色的甲瓒,在450 nm左右有最大吸收峰。通过450 nm下测定吸光值变化可测得体系中Gln含量。

这个试剂盒检测的是L-谷氨酰胺,检测不了L-丙氨酸-谷氨酰胺。L-丙氨酰谷氨酰胺在培养基中比较稳定,L-丙氨酰谷氨酰胺被细胞摄入之后,在细胞内可以转化成丙氨酸和谷氨酰胺。我们试剂盒检测的目标物质是L-谷氨酰胺,所以也就是说如果您培养基中添加的是L-丙氨酰谷氨酰胺这种物质,用我们试剂盒检测培养基样本,监测不到L-丙氨酰谷氨酰胺的变化情况。
