| 检测原理 | 本试剂盒采用双抗体夹心ELISA法。用抗人GLP-1抗体包被于酶标板上，实验时样品（或标准品）中的人GLP-1会与包被抗体结合。后依次加入生物素化的抗人GLP-1抗体和辣根过氧化物酶标记的亲和素，抗人GLP-1抗体与结合在包被抗体上的人GLP-1结合，生物素与亲和素特异性结合而形成免疫复合物，游离的成分被洗去。加入显色底物(TMB)，TMB在辣根过氧化物酶的催化下呈现蓝色，加终止液后变成黄色。用酶标仪在450 nm波长处测OD值，GLP-1浓度与OD450值之间呈正比，通过绘制标准曲线计算出样品中GLP-1的浓度。 |

胰高血糖素样肽-1 ( GLP-1 ) 是一种 30 或 31 个氨基酸长的肽激素，源自胰高血糖素原肽的组织特异性翻译后加工。初始产物 GLP-1 (1–37) 易于酰胺化和蛋白水解裂解，从而产生两种截短且等价的生物活性形式：GLP-1 (7–36) 和 GLP-1 (7–37) 。内源性 GLP-1 主要被二肽基肽酶 4 (DPP-4) 以及中性肽链内切酶 24.11 (NEP 24.11) 和肾脏清除快速降解，半衰期约为2 分钟。因此，只有 10-15% 的 GLP-1 完整地进入循环，导致空腹血浆水平仅为 0-15 pmol/L。
我们的GLP-1试剂盒针对完整GLP-1蛋白进行检测，没有办法检测其代谢产物
另外，有的客户为了防止GLP-1降解在会样本中加入格列汀(DPPIV抑制剂)和三氟乙酸，但我们并不建议添加有机试剂到样本中，这可能会对ELISA检测造成干扰。
