检测原理 | 本试剂盒采用双抗体夹心ELISA法。用抗人ICAM-1/CD54抗体包被于酶标板上,实验时样品(或标准品)中的人ICAM-1/CD54会与包被抗体结合。后依次加入生物素化的抗人ICAM-1/CD54抗体和辣根过氧化物酶标记的亲和素,抗人ICAM-1/CD54抗体与结合在包被抗体上的人ICAM-1/CD54结合,生物素与亲和素特异性结合而形成免疫复合物,游离的成分被洗去。加入显色底物(TMB),TMB在辣根过氧化物酶的催化下呈现蓝色,加终止液后变成黄色。用酶标仪在450 nm波长处测OD值,ICAM-1/CD54浓度与OD450值之间呈正比,通过绘制标准曲线计算出样品中ICAM-1/CD54的浓度。 |

Q1:ICAM-1和ICAM-4有什么区别呢?
ICAM蛋白家族由五个成员组成,分别是ICAM-1到ICAM-5。已知它们在炎症和免疫反应中与白细胞整合素CD11/CD18(如LFA-1和巨噬细胞-1抗原)结合。
ICAM-1又叫做CD54,在静息的血管内皮细胞(VEC)上呈低水平表达,通过与血管内皮细胞表面上的特异性受体结合而发挥其生物学活性。
ICAM-4 (LW血型抗原)表达于红细胞、红系前体细胞和其他血细胞 (包括 T 和 B 细胞)上,维持红细胞表面和血管内皮之间的密切接触,也在各种正常和病理状态中发挥作用。
