检测原理	本试剂盒采用双抗体夹心ELISA法。用抗人IFNα抗体包被于酶标板上，实验时样品（或标准品）中的人IFNα会与包被抗体结合。后依次加入生物素化的抗人IFNα抗体和辣根过氧化物酶标记的亲和素，抗人IFNα抗体与结合在包被抗体上的人IFNα结合，生物素与亲和素特异性结合而形成免疫复合物，游离的成分被洗去。加入显色底物(TMB)，TMB在辣根过氧化物酶的催化下呈现蓝色，加终止液后变成黄色。用酶标仪在450 nm波长处测OD值，IFNα浓度与OD450值之间呈正比，通过绘制标准曲线计算出样品中IFNα的浓度。

Q1:人干扰素α(IFNα)酶联免疫吸附测定试剂盒, 检测哪种亚型IFNα? 如果检测的是总的，那标准品是混合物吗？

该指标测得是所有亚型的IFNα，也就是总的IFNα。标准品并不是所有亚型的混合物，而是人源的IFN-alpha 2a （重组蛋白，表达体系：HEK293细胞）。但由于人的15种IFN-α亚型同源性高达80%，所以我们试剂盒中所用到的抗体可识别所有亚型，另外这款试剂盒对不同亚型的IFNα做过验证，均是可以检测到的。
