检测原理	本试剂盒采用双抗体夹心ELISA法。用抗人MMP-2抗体包被于酶标板上，实验时样品（或标准品）中的人MMP-2会与包被抗体结合。后依次加入生物素化的抗人MMP-2抗体和辣根过氧化物酶标记的亲和素，抗人MMP-2抗体与结合在包被抗体上的人MMP-2结合，生物素与亲和素特异性结合而形成免疫复合物，游离的成分被洗去。加入显色底物(TMB)，TMB在辣根过氧化物酶的催化下呈现蓝色，加终止液后变成黄色。用酶标仪在450 nm波长处测OD值，MMP-2浓度与OD450值之间呈正比，通过绘制标准曲线计算出样品中MMP-2的浓度。

Q1:ELISA可以测细胞上清液MMP-2和MMP-9吗？我看其他人写的论文，除了测这两个指标表达量，还要测活性，ELISA是不是可以测活性？跟明胶酶谱法有什么区别？

这两款试剂盒可以用于您的细胞上清的检测的。不过ELISA检测细胞上清受到培养基组分、细胞生长情况、以及外部药物刺激条件等因素的影响，建议您正式实验前先开展预实验。
明胶酶谱法是利用SDS与样品中的MMPs的可逆性结合，将样本中的MMP-2和MMP-9分离，再通过二价金属离子缓冲体系恢复两者的活性，该试验方法是专门针对MMP-2和MMP-9的活性检测的。而ELISA是通过抗原抗体的结合反应去检测待测物的含量（即浓度），即通过MMP-2和MMP-9的特异性抗体去捕获样本的MMP-2和MMP-9。两者的实验原理不同，ELISA无法检测MMP-2和MMP-9的活性。
