检测原理 |
HDL、CM 和 VLDL 等脂蛋白(除 LDL)首先在表面活性剂作用下改变结构并解离,所释放出来的微粒化胆固醇分子与胆固醇酶试剂反应,产生的过氧化氢在缺乏偶联剂时被消耗而不显色,此时 LDL 颗粒仍是完整的,再加入含有偶联剂的试剂,它可使 LDL 颗粒解离释放胆固醇,胆固醇在胆固醇酯酶(CE )、胆固醇氧化酶 ( CO ) 催化下,产生过氧化氢,过氧化氢在 4-氨基安替吡啉(4-AA)和酚(T-OOS)存在时,经过氧化物酶(POD)催化,反应生成苯醌亚胺非那腙的红色醌类化合物,因其他脂蛋白的胆固醇分子已除去,色泽深浅与 LDL-C 量呈比例。
