检测原理	本试剂盒采用双抗体夹心ELISA法。用抗小鼠IgA抗体包被于酶标板上，实验时样品（或标准品）中的小鼠IgA会与包被抗体结合。后依次加入生物素化的抗小鼠IgA抗体和辣根过氧化物酶标记的亲和素，抗小鼠IgA抗体与结合在包被抗体上的小鼠IgA结合，生物素与亲和素特异性结合而形成免疫复合物，游离的成分被洗去。加入显色底物(TMB)，TMB在辣根过氧化物酶的催化下呈现蓝色，加终止液后变成黄色。用酶标仪在450 nm波长处测OD值，IgA浓度与OD450值之间呈正比，通过绘制标准曲线计算出样品中IgA的浓度。

Q1:检测小鼠血清里的sIgA，会检测到IgA吗？血清样本中的IgA会干扰sIgA的检测吗？

样本中的IgA是可以检测到的，这主要是基于IgA和sIgA结构的重叠，会存在一定的交叉反应。
小鼠和人类的IgA生物学在几个方面有所不同。在人血清中，IgA主要以单体形式存在，而在小鼠血清中，IgA主要以聚合形式存在。此外，人类IgA可分为密切相关的IgAl和IgA2亚类，其中IgA2较不易被蛋白水解降解。在血清中，IgAl亚类占主导地位，而在分泌物中发现的主要亚型是IgA2，尽管IgAl和IgA2都可以检测到sIgA。
