检测原理 | 本试剂盒采用双抗体夹心ELISA法。用抗小鼠sIgA抗体包被于酶标板上,实验时样品(或标准品)中的小鼠sIgA会与包被抗体结合。后依次加入生物素化的抗小鼠sIgA抗体和辣根过氧化物酶标记的亲和素,抗小鼠sIgA抗体与结合在包被抗体上的小鼠sIgA结合,生物素与亲和素特异性结合而形成免疫复合物,游离的成分被洗去。加入显色底物(TMB),TMB在辣根过氧化物酶的催化下呈现蓝色,加终止液后变成黄色。用酶标仪在450 nm波长处测OD值,sIgA浓度与OD450值之间呈正比,通过绘制标准曲线计算出样品中sIgA的浓度。 |

样本中的IgA是可以检测到的,这主要是基于IgA和sIgA结构的重叠,会存在一定的交叉反应。
小鼠和人类的IgA生物学在几个方面有所不同。在人血清中,IgA主要以单体形式存在,而在小鼠血清中,IgA主要以聚合形式存在。此外,人类IgA可分为密切相关的IgAl和IgA2亚类,其中IgA2较不易被蛋白水解降解。在血清中,IgAl亚类占主导地位,而在分泌物中发现的主要亚型是IgA2,尽管IgAl和IgA2都可以检测到sIgA。
