检测原理 | 本试剂盒采用竞争ELISA法。用小鼠T抗原包被于酶标板上,实验时加入样品(或标准品)和辣根过氧化物酶(HRP)标记的抗T单克隆抗体,样本(或标准品中)的T与包被抗原T竞争结合HRP标记的抗T单抗上的结合位点,形成包被抗原T-HRP标记的抗T单抗复合物,游离的成分被洗去。加入显色底物(TMB),TMB在辣根过氧化物酶的催化下呈现蓝色,加终止液后变成黄色。用酶标仪在450 nm波长处测OD值,T浓度与OD450值之间呈反比,通过绘制标准曲线计算出样品中T的浓度。 |

含酚红的培养基可能对睾酮检测有影响。酚红在培养基中被用来作为PH值的指示剂:中性时为红色,酸性时为黄色,碱性时为紫色。研究表明,酚红可以模拟固醇类激素的作用(特别是雌激素)。为避免固醇类反应及对细胞的毒性作用,培养细胞,尤其是哺乳类细胞时,用不加酚红的培养基。
介绍一个小窍门:培养基在分装时,留一管加酚红,放到培养箱内平衡(可以看出PH值的变化,又可以看培养基是否污染)。在培养前放到培养箱内平衡再使用(可以避免培养时使用酚红)。
