| 检测原理 | 本试剂盒采用竞争ELISA法。用大鼠T抗原包被于酶标板上，实验时加入样品（或标准品）和辣根过氧化物酶（HRP）标记的抗T单克隆抗体，样本（或标准品中）的T与包被抗原T竞争结合HRP标记的抗T单抗上的结合位点，形成包被抗原T-HRP标记的抗T单抗复合物，游离的成分被洗去。加入显色底物(TMB)，TMB在辣根过氧化物酶的催化下呈现蓝色，加终止液后变成黄色。用酶标仪在450 nm波长处测OD值，T浓度与OD450值之间呈反比，通过绘制标准曲线计算出样品中T的浓度。 |

含酚红的培养基可能对睾酮检测有影响。酚红在培养基中被用来作为PH值的指示剂：中性时为红色，酸性时为黄色，碱性时为紫色。研究表明，酚红可以模拟固醇类激素的作用（特别是雌激素）。为避免固醇类反应及对细胞的毒性作用，培养细胞，尤其是哺乳类细胞时，用不加酚红的培养基。
介绍一个小窍门：培养基在分装时，留一管加酚红，放到培养箱内平衡（可以看出PH值的变化，又可以看培养基是否污染）。在培养前放到培养箱内平衡再使用（可以避免培养时使用酚红）。
