| 检测原理 | 本试剂盒采用双抗体夹心ELISA法。用抗大鼠Bcl-2抗体包被于酶标板上，实验时样品（或标准品）中的大鼠Bcl-2会与包被抗体结合。后依次加入生物素化的抗大鼠Bcl-2抗体和辣根过氧化物酶标记的亲和素，抗大鼠Bcl-2抗体与结合在包被抗体上的大鼠Bcl-2结合，生物素与亲和素特异性结合而形成免疫复合物，游离的成分被洗去。加入显色底物(TMB)，TMB在辣根过氧化物酶的催化下呈现蓝色，加终止液后变成黄色。用酶标仪在450 nm波长处测OD值，Bcl-2浓度与OD450值之间呈正比，通过绘制标准曲线计算出样品中Bcl-2的浓度。 |

Q21:ELISA的孵育过程是否可以用细胞培养箱进行实验？
可以使用，但是细胞培养是需要无菌环境的，ELISA不需要无菌，我们建议客户尽量不要用这个仪器，避免做其他实验如细胞培养时造成污染，可以选择常规的烘箱。
Q22:ELISA样本分组中Mock Control Vector的区别
mock更接近于negative control（NC）就是空白对照，啥都不加。 control是对照，比如加药，光加药物溶剂（DMSO）的叫control vector一般是人工构建的过表达或RNAi载体去转染细胞时，光转染空白质粒那一组叫vector，理论上，也是对照的一种，表明你转染的质粒本身对细胞的某个生理现象无影响。

ELISA未包被试剂盒是不能出售单组分的（没有对应的单组分货号），不能出售，推荐客户购买整套试剂盒。
Q24:ELISA抗凝剂中肝素是否可以使用？
ELISA推荐使用EDTA作为抗凝剂，尽量避免肝素作为抗凝剂(过量肝素可能造成检测结果偏高,若使用肝素抗凝剂，请确保肝素浓度低于10 IU /mL
Q25:ELISA实验中，孵育阶段可以使用摇床或是水浴锅进行孵育吗？
为了保证孵育阶段温度波动控制在1℃以内，推荐客户使用恒温箱进行孵育
如果确实因为条件限制无法使用恒温箱孵育，可以考虑使用水浴锅孵育，但需要注意避免酶标板被水浴用水污染
不建议使用摇床进行孵育，尤其是带转速的摇床
Q26:ELISA实验中加入终止液终止显色反应的原理是什么？
加入终止液后，一方面，破坏了 HRP酶的活性，使酶的催化功能丧失；另一方面， 由于pH改变而使得最终的颜色发生变化。
