| 检测原理 |
DCFH-DA（2,7-dichlorofuorescin diacetate）是一种可以自由穿过细胞膜的荧光探针，自身没有荧光，进入细胞内后，可以被细胞内的酯酶水解生成DCFH（dichlorofluorescin）。在活性氧存在时DCFH被氧化为不能透过细胞膜的强绿色荧光物质DCF（dichlorofluorescein），其荧光在激发波长502nm，发射波长530nm附近有最大波峰，强度与细胞内活性氧水平成正比。

该试剂盒适用于检测活细胞ROS，组织匀浆处理后，细胞已经破碎死亡，因此该试剂盒不能检测组织匀浆。如果是新鲜组织样本，制备成单细胞悬液，可以检测。

该ROS试剂盒检测的ROS没有特指针对某一类ROS，检测的是总的ROS，因为探针的特异性没有那么强。

ROS试剂盒检测的是强绿色荧光，如果样本自身携带的荧光也是绿色的就有干扰，其他颜色的荧光对该试剂盒检测没有影响。
