检测原理 | 本试剂盒采用竞争ELISA法。用ST/5-HT抗原包被于酶标板上,实验时样品或标准品中的ST/5-HT与包被的ST/5-HT竞争生物素标记的抗ST/5-HT单抗上的结合位点,游离的成分被洗去。加入辣根过氧化物酶标记的亲和素,生物素与亲和素特异性结合而形成免疫复合物,游离的成分被洗去。加入显色底物(TMB),TMB在辣根过氧化物酶的催化下呈现蓝色,加终止液后变成黄色。用酶标仪在450nm波长处测OD值,ST/5-HT浓度与OD450值之间呈反比,通过绘制标准曲线计算出样品中ST/5-HT的浓度。 |

Q21:ELISA的孵育过程是否可以用细胞培养箱进行实验?
可以使用,但是细胞培养是需要无菌环境的,ELISA不需要无菌,我们建议客户尽量不要用这个仪器,避免做其他实验如细胞培养时造成污染,可以选择常规的烘箱。
Q22:ELISA样本分组中Mock Control Vector的区别
mock更接近于negative control(NC)就是空白对照,啥都不加。 control是对照,比如加药,光加药物溶剂(DMSO)的叫control vector一般是人工构建的过表达或RNAi载体去转染细胞时,光转染空白质粒那一组叫vector,理论上,也是对照的一种,表明你转染的质粒本身对细胞的某个生理现象无影响。
Q23:ELISA未包被试剂盒能不能出售单组分?
ELISA未包被试剂盒是不能出售单组分的(没有对应的单组分货号),不能出售,推荐客户购买整套试剂盒。
Q24:ELISA抗凝剂中肝素是否可以使用?
ELISA推荐使用EDTA作为抗凝剂,尽量避免肝素作为抗凝剂(过量肝素可能造成检测结果偏高,若使用肝素抗凝剂,请确保肝素浓度低于10 IU /mL
Q25:ELISA实验中,孵育阶段可以使用摇床或是水浴锅进行孵育吗?
为了保证孵育阶段温度波动控制在1℃以内,推荐客户使用恒温箱进行孵育
如果确实因为条件限制无法使用恒温箱孵育,可以考虑使用水浴锅孵育,但需要注意避免酶标板被水浴用水污染
不建议使用摇床进行孵育,尤其是带转速的摇床
Q26:ELISA实验中加入终止液终止显色反应的原理是什么?
加入终止液后,一方面,破坏了 HRP酶的活性,使酶的催化功能丧失;另一方面, 由于pH改变而使得最终的颜色发生变化。
