检测原理 |
通过黄嘌呤及黄嘌呤氧化酶反应系统产生超氧阴离子自由基(O2-),它氧化羟胺形成亚硝酸盐,在显色剂作用下产生紫红色化合物。当反应体系中加入SOD后,催化超氧阴离子自由基发生歧化反应,使形成的亚硝酸盐减少,进而产生的紫红色变浅。根据样品管与对照管中的吸光度差值,来计算SOD的活性。同时,经过试剂处理的样本导致Mn-SOD失活,而不影响CuZn-SOD。
