检测原理 |
GSH-Px可以促进过氧化氢(H2O2)与还原型谷胱甘肽(GSH)反应生成H2O及氧化型谷胱甘肽(GSSG),反应式见下图。通过测定底物GSH在单位时间内被氧化的速度来表示酶的活力,反应剩余的GSH与二硫代双二硝基苯甲酸(DTNB)作用生成的黄色硫代硝基苯甲酸阴离子(TNB),测定该阴离子的浓度来计算出GSH的减少量。由于GSH在没有酶的条件下也能进行氧化(非酶反应),所以最后计算此酶活力时,必须扣除非酶反应引起的GSH减少的那部分。
