Q1:该如何选择二抗?
二抗根据一抗的来源和客户的实验需求来选择。
先看实验所用一抗的种属来源和亚型。根据一抗种属来源和亚型来决定二抗:如一抗是兔来源的IgG,那就选抗兔IgG的二抗,例如Goat Anti-Rabbit IgG;如一抗是小鼠来源的IgM,那就选抗小鼠IgM的二抗,例如Goat Anti-Mouse IgM。
标记物的选择。常见的标记物有有HRP、Biotin、荧光素等。
ELISA常用Biotin标记的二抗,后接一个链霉亲和素标记的HRP起到进一步信号放大作用,用TMB底物显色;
WB主要选择HRP标记的二抗,用ECL底物发光液显色;
IHC一般也是选择HRP标记的二抗,推荐使用免疫组化试剂盒(E-IR-R217或最新的E-IR-R220/221),一般用DAB底物液显色;
IF可按实验需要选择不同荧光素标记的二抗,如FITC、Cy3、PE等均可(尽量避开细胞的自发荧光),通过荧光显微镜检测荧光。
