线粒体呼吸链复合物Ⅱ又称琥珀酸-辅酶Q还原酶，广泛存在于动物、植物、微生物和培养细胞的线粒体中，它催化三羧酸循环中的琥珀酸氧化成延胡索酸（富马酸) ，通过复合物中的基本单元结构比如硫铁蛋白等，使泛醌转化为还原形态。复合体Ⅱ的催化产物还原型辅酶Q可进一步还原 2,6-二氯吲哚酚，2,6-二氯吲哚酚在600 nm有特征吸收峰，通过检测2,6-二氯吲哚酚的减少速率来计算该酶活性。
