Q1:细胞染色缓冲液有什么用，我可以用PBS代替吗？一个样本需要多少细胞染色缓冲液？
细胞染色缓冲液主要作用可以保持细胞活力，使细胞变得稳定并且对损伤不太敏感；使pH敏感荧光染料产生的荧光信号强度最大化；减少抗体与靶细胞的非特异性结合；螯合金属阳离子，减少细胞之间的粘连。可以用1%BSA的PBS溶液替代。一个样本大概需要1.5 mL细胞染色缓冲液。
Q2:我的样本是小鼠的外周血和肺泡灌洗液，我取出之后怎么样能最大时间的保存样本？
流式实验建议用新鲜细胞做。如果您这边情况特殊，只能建议您先将制备好的单细胞悬液用4%多聚甲醛固定30min，然后用PBS洗涤两遍，保存在细胞染色缓冲液（可以用1 %BSA的PBS替代）中，置于4°C保存，试剂收到后尽快检测。
