检测原理	细胞增殖检测广泛应用于细胞活性、遗传毒性及抗肿瘤药物效果的评价中，直接检测细胞中的DNA合成被认为是最精确的检测细胞增殖的方法。最初广泛使用的检测细胞中DNA合成的方法是放射性标记核苷掺入法，但该方法由于有放射性污染并且很难实现单细胞检测而受到很大的限制，随后逐渐被基于抗体检测的BrdU法所替代。BrdU法步骤繁多，且需要使用BrdU抗体，影响因素较多，稳定性较差。EdU法基于EdU掺入和后续的点击反应，无需使用抗体、操作便捷、检测灵敏度高，是一种在BrdU法基础上升级换代的新方法，将会逐步取代BrdU法。EdU(5-ethynyl-2-deoxyuridine)，中文名5-乙炔基-2-脱氧尿苷，是胸腺嘧啶脱氧核苷类似物，EdU可以在DNA合成过程中替代胸腺嘧啶脱氧核苷掺入到新合成的DNA中。另一方面，EdU上的乙炔基能与荧光标记的小分子叠氮化物探针（如FITC Azide、Elab Fluor® 488 Azide、Elab Fluor® 594 Azide、Elab Fluor® 647 Azide），通过一价铜离子的催化发生共价反应，形成稳定的三唑环，该反应非常迅速，被称作点击反应（Click reaction）。通过点击反应，新合成的DNA会被相应的荧光探针所标记，从而可以使用适当的荧光检测设备检测到增殖的细胞。
