| 检测原理 | MTT是噻唑蓝（3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide），可被线粒体内的一些脱氢酶还原生成结晶状的深紫色产物甲臜（formazan）。甲臜可被DMSO完全溶解，并通过酶标仪测定570nm波长附近的吸光度。细胞增殖速度越快，则吸光度越高；细胞毒性越大，则细胞增殖减慢从而导致吸光度降低。 |

Q1:请问CCK8和凋亡的摸出来的药物IC50不一样的原因。
IC50指的是半抑制率，可以用CCK8检测，凋亡实验检测的是凋亡率，这个不等同于IC50。这两个实验的实验目的是不一样的。
Q2:MTS, XXT, WST-1, CCK-8（WST-8）检测试剂盒的差异？
MTT检测试剂盒广泛用于检测细胞增殖和细胞毒性，而MTS, XXT, WST-1, WST-8都是MTT的升级替代产品，和MTT相比有明显优点。首先，MTT被线粒体内的一些脱氢酶还原成生成的formazan不是水溶性的，需要有特定的溶剂来溶解。WST-8和XTT，MTS产生的formazan都是水溶性的，可以省去后续的溶解步骤。其次，WST-8产生的formazan比XTT，MTS产生的formazan更易溶解。最后，WST-8比MTT，XTT更稳定，线性范围更宽，灵敏度更高。
